Why Empore™

Why Labs Choose Empore StageTips

Pre-packed, standardized, and ready to use — no more manual tip packing.

1

Pre-Packed — No Manual Preparation

Traditional StageTips require manually punching membrane disks with a syringe needle, placing them into a pipette tip, and pressing into place. Empore™ StageTips come pre-packed — open the pack and start your experiment.

2

Excellent Lot-to-Lot Reproducibility

QA/QC testing across multiple lots shows no significant variation (p > 0.05) in protein identification and PSM counts. Pearson r > 0.93 for quantified proteins between different tips and lots.

3

9 Sorbent Chemistries, Up to 6 Layers

C18, C8, SDB-XC, SDB-RPS, SAX, SCX, HLB, and pre-configured multi-layer combinations (C18/SCX, C18/SCX/C18). Stack layers for integrated desalting and fractionation in a single tip.

4

Automatable — Hands-Free Processing

Use our rack adaptors to process StageTips by centrifugation — hands-free, scalable, and high throughput. No manual pushing. No syringe needed. View Rack Adaptors →

PROTOCOL

C18 Desalting in 5 Steps

Complete peptide cleanup in under 30 minutes with our validated 5-step protocol - from sample preparation to MS-ready peptides

StageTip C18 Desalting Step Snapshot_edited.png

1

Conditioning

  1. Filter through 50 – 100 µL of methanol without letting all solution pass, keeping the membrane wet. Discard filtrate.

  2. Repeat conditioning if membrane has gone dry.

  3. For centrifugation processing we recommend 1,000 – 1,500 x g for all steps.

2

Equilibrating

  1. Filter through 50 – 100 µL of 0.5% trifluoroacetic acid without letting all solution pass, keeping the membrane wet. Discard filtrate.

  2. Restart protocol at conditioning if membrane has gone dry.

3

Sample loading

  1. Put StageTip into a new collection tube and pipette sample into StageTip. Maximum volume of 400 µL. 40 µg maximum recommended peptide mass per C18-HD StageTip.

  2. Pass all solution through membrane and keep filtrate for troubleshooting problems.

  3. Optionally, user may pass the sample filtrate through an additional time, which may improve retention.

4

Washing

  1. Put StageTip into a new collection tube.

  2. Filter through 50 – 100 µL of 0.5% trifluoroacetic acid.

  3. Filter through 50 – 100 µL of 0.5% acetic acid.

  4. Depending on the level of contaminants in the sample, more wash steps may be necessary.

5

Sample eluting

  1. Put StageTip into a new collection tube for your analyte.

  2. Filter through 10 – 30 µL of 80% acetonitrile, 0.5% acetic acid, in water.

  3. Repeat step 5b one to two more times to elute all the analyte.

  4. Elution volume for high recovery may depend on how many micrograms of sample loaded. At maximum capacity, 100 µL total over a few elutions may be necessary.

  5. The sample is clean and ready for further concentration followed by mass spectrometry analysis.
