Protocol
1. Conditioning: Pass 50 µL methanol through the tip, then 50 µL of 0.2% TFA.
2. Sample loading: Load acidified sample (pH < 3) onto the membrane. Pass through twice for best retention.
3. Wash: Wash with 50–100 µL of 0.2% TFA, followed by 50–100 µL of 0.5% acetic acid (AcOH).
4. Elute: Elute with 10–30 µL of 80% acetonitrile / 5% NH4OH (single fraction) or use 100 mM HCOONH4 / 40% & 60% ACN for two-fraction high-pH separation. Repeat 1–2 times for complete recovery.
